A very large anaconda captured from the Amazon River basin is being kept in a scientific lab owned Wexel Hall for experimentation. The research project is lead by Dr Amanda Hayes (Crystal Allen) and funded by Murdoch (John Rhys-Davies), a well-known industrialist. While visiting the facility, Murdoch agitates the queen anaconda with a torch, and she is gassed to calm her down. However, as Murdoch, his assistant Pinkus (Ryan McCluskey), and a scientist leave, the anaconda beaks through her enclosure's wall. As it escapes, it kills many of the people working in the lab. In looking through her enclosure, Amanda and another researcher realize that a second anaconda has also escaped. Murdoch calls in a team of animal hunters, led by Hammett (David Hasselhoff), to capture both snakes. Amanda and Pinkus go with them. At a small farm in the middle of the woods, the owner is eaten alive by one of the snakes. The hunting party arrives later and begin formulating a plan of attack. While confronting the snake, four of the party are killed.
As they regroup, Amanda reluctantly reveals that the queen snake is pregnant and will give birth in less than 24 hours. The teams wants to call in the military, but Hammett forbids it and threatens Amanda with jail time for her role in creating it. In the morning, the party begins searching for the snake, but Pinkus is killed. While Hammett searches for it on foot, Amanda and one of the two remaining hunters search in the jeep. They spot the snake going into an old factory, and follow her in to plant explosives around the building. Before they can finish, queen attacks. The hunter helps Amanda escape but is captured himself. Before the snake kills him, he is able to discharge a grenade, killing himself and the snake. Hammett and his last remaining hunter arrive and hear the grenade go off. The hunter moves to call the military, so Hammett kills him. Inside the factory, Amanda realizes Hammett is working for Murdoch and tries to avoid them. Inside the building, the queen gives birth. After wounding Hammett, Amanda escapes from the building leaving him to be attacked by the baby snakes. Once she is at a safe distance, she detonates the explosives. As she sits by a roadside burning all of her documentation on the snake research, one of Murdoch's men drives by heading to the factory. Inside he finds one baby snake alive and captures it to take back to Murdoch.
